need help looking to drill my 50 gallon for rear bulkheads what is the best place to drill the tank for the drain and return is it one inch from the top and side and should they both be equal in hight?

How many and what size? You at least want to stay a hole size away from the edges.

im using a 1 inch for the return and 3/8 for the fill i want to put them 1 1/2 from the top and sides of the tank return on the right and drain on the left?

be carful drilling holes close together - especially for a tank that's 3/16 or 1/4" glass. I would have the feed on one side and the drain on the other.
