Building a stand for a reef ready 125 gallon tank with to overflows.
1 - should i use 4x4 with two 2x2 for the the legs.
2 - can i only put a center 2 2x2 only in back keepin the front open.
3 - can u use 2x6 for the top

So many easy ways to make it. Check DIY here and reef central. Even you tube it too. You can get ideas. I would use 2x6 for top and bottom . Use 2x4 for legs part. I would make the stand bigger so you can cut hole for your reef ready on both sides. If you don't you will run in to cutting part of structure of legs. Just remember where holes would line up when you make length and width. Do research that would help you better understand so you can see how you want it. People even post pictures on doing it. This is easy and fun to do. If you want when I brought my wood at Home depot I gave them the measurements what I needed. They did all cutting. All I did buy screws and put it together.
