I would use either a hole saw, using light pressure as not to break anything or a dremel tool with a carbide grinding bit. If you go with the dremel, mask of the area with masking tape, mark the hole and have at it. Also with the dremel, too high of a speed and the acrylic will melt onto the bit, so don't use the highest speed, about medium speed should work.

put masking tape on both side of plexeglass and use a hole saw little pressure on drill and not to fast of speed let the holesaw do the cuting, before you go thur one side use holesaw on other side that way will a clean cut on both side
