I also take the calerpa out of my refugium on a weekly basis, but I usually have more than a cup of it, so I put it in a large bucket, add some tank water and shake out the critters (there are a lot hiding in there), then add them to the main tank with the water.

You really need to get a copy of Joyce Wilkerson's book "Clownfishes". It will answer all of your questions so far and another hundred that you haven't thought of yet. She also discusses how to raise clownfish, if that is your ultimate goal. Good luck!
